五指毛桃组培育苗研究

摘要:
以五指毛桃 Ficus hirta 优良植株健壮的带顶芽嫩枝为外植体,进行组织培养育苗研究,结果表明:最合适的外植体灭菌方法是1 g·L-1 的升汞溶液灭菌14 min;合适的芽诱导培养基为MS+6-BA 1.0 mg·L-1+IBA 0.1 mg·L-1+ 蔗糖 30 g·L-1+卡拉胶6.5 g·L-1,诱导率为40%;最适的增殖培养基为MS+6-BA 1.0 mg·L-1+IBA 0.3 mg·L-1+ 蔗糖30 g·L-1+卡拉胶6.5 g·L-1,增殖倍数为5.43;最佳的生根培养基为1/4MS+IBA 1.5 mg·L-1+NAA 0.3 mg·L-1+蔗糖20 g·L-1+卡拉胶6.5 g·L-1+活性碳0.5 g·L-1,生根率为96%;最佳的瓶苗移栽基质为轻型基质,存活率为98.15%。

Study on Tissue Culture of Ficus hirta

Abstract:
The tissue culture of Ficus hirta was studied, selecting the robust shoots with apical buds of the as experiment material. The results showed that in this study the most suitable sterilization treatment for shoots is soaking in 1 g·L-1 mercury solution for 14 minutes. The most suitable bud induction medium is MS+6-BA 1.0 mg·L-1+IBA0.1 mg·L-1+sucrose 30 g·L-1+carrageenan 6.5 g· L-1, which induce rate is 40%. The most suitable proliferation medium is MS+6-BA 1.0 mg·L-1+IBA0.3 mg · L-1+sucrose 30 g·L-1+carrageenan 6.5 g·L-1, which the multiplication index is 5.43. The best rooting medium is 1/4MS+IBA1.5 mg·L-1+NAA0.3 mg·L-1+sucrose 20 g·L-1+carrageenan 6.5 g·L-1 + activated carbon 0.5 g·L-1, which the rooting rate is 96%. The best seedling transplanting medium is a light substrate with a survival rate of 98.15%.
